Stephen Evanko: Stephen Evanko received his M.S. and Ph.D. degrees from the University of New Mexico in 1990 and 1993, respectively. His work focused on the response of connective tissues to alterations in mechanical loading and the role of proteoglycans and hyaluronan in the material properties of tendon and fibrocartilage. He then joined the laboratory of Dr. Thomas Wight in the Department of Pathology, University of Washington, as a postdoctoral fellow, studying the role of hyaluronan and proteoglycans in smooth muscle cell proliferation and migration. He is currently a research fellow in Cardiovascular Research at The Hope Heart Institute (Seattle, WA), where he continues to study hyaluronan in the biology of the artery wall and cardiovascular disease.

Abnormal accumulation of hyaluronan matrix diminishes contact inhibition of cell growth and promotes cell migration.
Elevated hyaluronan biosynthesis and matrix deposition correlates with cell proliferation and migration. We ectopically expressed three isoforms of hyaluronan synthase (HAS1, HAS2, or HAS3) in nontransformed rat 3Y1 cells and observed a de novo, massive formation of a hyaluronan matrix that resulted in a partial loss of contact-mediated inhibition of cell growth and migration. All three HAS tra...
متن کاملThe human hyaluronan receptor RHAMM is expressed as an intracellular protein in breast cancer cells.
The receptor for hyaluronan mediated motility (RHAMM) has been reported to mediate migration, transformation, and metastatic spread of murine fibroblasts. Here we describe the expression of two human RHAMM isoforms, which are generated by alternative splicing of the primary gene transcript, by a series of human breast carcinoma cell lines. A polyclonal antibody, raised against a bacterially exp...
متن کاملIdentification of IHABP, a 95 kDa intracellular hyaluronate binding protein.
The extracellular matrix component hyaluronan is believed to play important roles in various processes of organogenesis, cell migration and cancer. Recognition of and binding to hyaluronan is mediated by cell surface receptors. Three of them, CD44, ICAM-1 and RHAMM (receptor for hyaluronic acid mediated motility), have been identified. A cDNA clone designated RHAMM turned out to possess transfo...
متن کاملInternalization of hyaluronan by chondrocytes occurs via receptor-mediated endocytosis.
Several studies have suggested that chondrocytes must have the capacity to internalize and degrade extracellular hyaluronan. In the present study we show direct evidence that hyaluronan is, in fact, endocytosed by chondrocytes and that the endocytosis is mediated via cell surface CD44/hyaluronan receptors. Cultures of bovine articular chondrocytes as well as rat chondrosarcoma chondrocytes were...
